You don't need to harvest them. I never needed to. I would save them. If you attack them, the BigDaddy's attack you and they can be a pain in the ass.

And it's some much more fun to shoot at big monsters then looking at them.

Depending on the situation, if I have the equipment nessercary to take down a "BigDaddy" then a little sisters is going to have her head blown in, if not, I play the hero and rescue her because I don't need to have my chest drilled out, thank you very much.

I beat the game, and I can tell you this. Whether you harvest them or save them effects the ending cinematic you get, so do whatever you want. Also if you save them you get much more benefit out of it cause you get rewarded later on. :]

I'd rescue. After all, you want to uncover the history, not go with what's going on....

The fact that you get more Adam from harvesting is false. Once you rescue like 3 Little Sisters, you get a reward that indeed contains Adam and a rare Plasmid.

I havn't gotten far enough in the game to have the choice, but I would rescue.
